`Slat—inns
`
`. A method for selecting a DNA sequence with a gene transcription
`
`modulating quality, comprising providing a transcription system with a
`
`variety of fragment-comprising vectors, said vectors further comprising i)
`
`an element with a gene—transcription repressing quality, and ii) a promoter
`
`directing transcription of a reporter gene, the method further comprising
`
`performing a selection step in said transcription system in order to identify
`
`a fragment comprising said DNA sequence with said gene transcription
`
`modulating quality.
`
`. A method according to claim 1, wherein said DNA sequence comprises a
`
`stable gene transcription modulating quality.
`
`. A method according to claim 1 or claim 2, wherein said DNA sequence
`
`comprises a gene transcription enhancing quality.
`
`. A method according to any one of claims 1—3, wherein said transcription
`
`system comprises host cells.
`
`. A method according to any one of claims 1-4, wherein said promoter may be
`
`active in said transcription system but wherein gene-transcription
`
`repression in said vectors results in gene-transcription repressing
`
`chromatin.
`
`. A method according to any of the preceding claims, wherein the DNA
`
`sequence involved in gene-transcription repression is a DNA sequence that
`
`is recognized by a protein complex and wherein said transcription system
`
`comprises said complex.
`
`. A method according to claim 5, wherein said complex comprises a
`
`heterochromatin-binding protein HPl, a Polycomb-group (PC-Gr) protein, a
`
`histone deacetylase activity or MeCP2 (methyl-CpG-binding protein.
