`cooling keep cycling rates high, and
`temperature accuracy is ensured
`through software algorithms. A

`tion configuration model (HLDHP).
`The PowcrBlock I (single block) and H (dual block)
`offer Pelrier heating and a small footprint. Two program-
`ming modes facilitate ease of use for simple (“Basic”) and
`advanced (“Linl<") applications. The singlc—blocl: model
`can be upgraded to dual blocks. Heated lids are optional.
`The water-cooled Singlefilock and DualBlock systems
`are the workhorses of Ericomp's line, with the flexibility to
`accommodate 0.5- and O.2—ml
`tubes, microplates, and
`slides. The low-cost systems featurc NTST traceable accu-
`racy, a five—year warranty, and across—the-block tempera-
`ture uniformity of :0.2°C. Optional items include heated
`lids and various block configurations.
`
`G-enex Lid. The small, uncomplicated Pacer cycler from
`Genex comes with a start and stop button and three LEDS
`to indicate cycling stages. This simple configuration is pos-
`sible because all of the programming and storage is per-
`formed on an accessory PC with the proprietary Preceptor
`software and then transferred to the machine before each
`run. Program storage space is limited, therefore, by the
`size of the computer rather than the cycler’s on—board
`memory. The Pcltier-cooled block can hold 16 05-1111 or
`20 0.2-ml tubes, two regular slides, or a mini 96-well plate.
`
`GL Applied Research The Genetic Thermal Cyclcr
`GTC-2 features two- and three—stcp amplification ability,
`built»in programmable autoextension, storage of up to 99
`protocols, and a 4-8-place sample block, among other fea-
`tures. Available options include a heated lid, a 96-well plate
`adapter, an 89-place sample block. that can hold up to 48
`D,5—ml and -‘l-l 0.Z~ml tubes, and a low—ternpcratI.u'c module.
`
`Hyba.-‘d Hybaid has just launched its Multibloclt system to
`complement
`three already-established, stylish,
`individual
`cyclers that fit a variety ofneeds. All ofits machines are PCR
`licensed, Pclticr cooled, and calibrated to NTST standards.
`The attractive PCR Sprint is claimed to be the lowest-
`priced licensed model available according to company lit-
`eratnre. It features fast ramping of up to 3°C/second,
`temperature control from 4—99°C, user-friendly program-
`ming, interchangeable blocks for 0.5 and 0.2 ml tubes, and
`a rli."ce—_vear guarantee. Hybaid’s PCR Express speeds up
`reactions involving its AGS Gold DNA polymerase with a
`onc»secon(l dwell time. Five control modes allow protocol
`flexibility—activc/simulated tube, simulated slide, and
`simulated block. An autoadjusting heated lid allows use of
`any plasticware and oil—f1-ee PCR in tubes and plates.
`Alphanumeric programming and advanced editing features
`simplify protocols, and four interchangeable blocks allow
`use of 0.2-rnl, 0.5—ml, .flat—l)l0cl(, and 38-l-well formats.
`The Ornn—F. cyclcr is a compact, multiformat, portable
`reactor with the ability to acconunodate 0.2-ml, 0.3-ml,
`and 0.5--ml tubes as well as 96-well plates. Active tube
`control and dynamic uniformity of 205°C across the plate
`allow precision and reproducibility between samples and
`experiments. An optional heated lid is available for oil-
`free cycling.
`In rim PCR protocols can be performed en masse with
`the Omnislide system, allowing simultaneous processing of

`Cepheid The SrnartCycler'" uses a unique cooling/he-ar-
`ing/optics/reaction module (the LCOREW) to perform
`amplification reactions with real-time monitoring. The
`company is working on modules that will handle all phas»
`es of sample preparation, reaction, and analysis, reducing
`the possibility of contamination.
`Corbett Research The new Rotor-Gene Real-Time
`Thermal Cyclet has two separate light sources to prevent
`spectral overlap in dual fluorescence studies. Samples can
`be loaded with reagents in the cap and reaction mix in the
`bottom of the tube. Centrifugal action mixes the samples
`and allows temperature uniformity. A 96-well adapter for
`use with Gcne—Disks will soon be available. Corbett also
`manufactures a Gradient ‘)6 VVell Cycler for microplatcs
`and a High Speed Capillary Cyclcr, as well as Cooled and
`Ambient 96 VVell Cyclers.
`
`Eppendor! Scientific Instruments The Mastercycler®
`series is sold with the PCR license. The company's gradi-
`ent model was the first on the market and offers a 96-
`position universal block that can accept 96 0.2-ml tubes, 77
`0.5-ml tubes, or one 96-well plate. Functions inc-udc selec-
`table gradients (from 1 to 20°C), Peltier heating, adjustable
`ramping, time extension, touchdown, program linking, and
`pause. A heated lid is
`included. According to Vincent
`Prezioso, product manager at Eppendorf Scientific,
`the
`uniform-rate cooling aspect allows users to accurately
`reproduce optimized conditions for each cycling pattern.
`The standard version Mastcrcycler includes all of the
`same features as the gradient model (without the gradient).
`The Mastercycler personal has a 25—posi'r_ion universal
`block and offers most of the features on the standard
`model. Personal memory cards allow users to program and
`retain their own cycle profiles and transfer them between
`any of the Mastcrcyclcr workstations.
`
`Ericomp The DeltaCyclcr T (single block) and H (dual
`block) systems are compact in design and use thermoelec-
`tric heating and cooling to minimize the accessory tubing
`that water—coolcd models require. A range of 4-“C to I 10°C
`with a temperature uniformity of :0.2°C allows the full
`range of protocols to be used. Options include two types of
`
`S 0 much has changed in the last l1alf—decadc in the
`
`world of gene ainplification. New breakthroughs and
`protocols have exceeded scientists’ wildest expecta-
`tions for applications of this essential technology. Is there a
`single life science lab in the country without a thermal
`cycler? From the high—end, ultIafast light—based cyclers to
`the tiny personal cyclers, there is a cyclcr to fit every need.
`VVay hack in the early years of PCR, “doing an amplifi-
`cation” meant simply that
`increasing the concentration of
`some target sequence of interest
`to use in insertion/
`ligation reactions. Quickly enough, scientists developed new
`ways to use the cycler. Now PCR-based methods exist for
`every aspect of biology that relies on genetic information.
`Forensic, genomic, and evolutionary studies have ben-
`efited greatly from the technique known as random ampli-
`fied polymorphic DNA (RAPD) PCR, which is used to
`fingerprint species from genomic DNA without prior
`sequence knowledge. Using arbitrary oligos, two Cycles of
`PCR with low soingcnty are followed by formal high-
`stringency PCR with specific primers. The reaction is then
`followed by RFLP or comparative analysis.
`Touchdown PCR was developed to increase the yield of
`specific products. in this method, the annealing tempera-
`ture is lowered one degree every second cycle to a lower
`limit or “touchdown" temperature. This touchdown tem-
`perature is then used for 10 cycles. This process ostensibly
`enriches for a specific product by taking advantage of dif-
`ferences in melting temperatures between correct and
`incorrect products by twofold per cycle.
`ll/iore recently,
`this technique has been adapted for genomic sequencing to
`elucidate DNA sequences from known peptide sequences.
`Long PCR is a method that allows amplification of
`long stretches of DNA, from 0.5 to 20 kb. An enzyme mix
`containing both proofreading and polymerase activities
`allows specific extension over great distances. Wl'1c11 used
`as an alternative to RFLP analysis, it shortens the process
`from six to eight weeks down to hours or days. It also may
`be used as an alternative to variable number of tandem
`repeats (VNTR) analysis.
`Chromosomal analysis has also benefited from PCR.
`DOP-PCR is a method that allows uniform amplification
`of sequences across a genome. Comparative DOP-PCR is
`used to detect chromosomal imbalances in genetic diseases
`and evolutionary studies. Amplified fragment length poly-
`morphism-PCR (AFLP-PCR) is used to detect allelic loss
`in aging and other studies. Single nucleotide polymor-
`phism (SNP) detection has made great strides with PCR.
`For hybrid cell studies researchers have developed Alu-
`PCR to detect human chromosomes in the hybrid cells.
`Scientists involved in the Human Genome Project
`have made great use of the cycle sequencing method.
`Developed as an alternative to the Sanger method, PCR-
`based sequencing uses femtomolar amounts of primers to
`start
`the elongation PCR,
`followed by sequencing. A
`major advantage is that this method can be used for linear
`double—stranded DNA, unlil<c the Sanger method. lt can
`also be used for cosmid, plaque, and colony DNA
`sequencing.
`Scientists studying RNA cxprcssion have not been left
`out of the PCR craze. Using reverse transcriptasc to pro»
`duce CDNA gave rise to RT—PCR. This method has been
`used to diagnose a variety ofinfectious and genetic diseases.
`Histologists quickly realized the advantage of having
`precise control over temperature and incubation time and
`adapted the cycler for use with 2'72 situ hybridization. One
`limitation oftraditional ISH is detection of low-copy num»
`her RNAs. This was overcome by the development of
`cycling—primed it: rim (cycling PRINS) labeling, which
`allowed researchers to gain deeper insight into the more
`ephemeral processes of the cell.
`PE Biosystems holds the exclusive license rights to the
`PCR technology. Anyone who wishes to perform automat-
`ed PCR using certain reagents and machines must first
`obtain a license. Fortunately, many reagents and machines
`include these rights in the purchase price, but the user
`must check with the supplier before performing the ampli-
`fication. This applies to research as well as applied fields. If
`the license is not supplied, PE Biosystems will provide one
`for a fee.
`This year’s field of cyclers includes many familiar
`names as well as some ingenious new models.
`
`Bio-Rad Bio—Rad’s newest model is the iCyclcr”‘, which
`boasts NIST accuracy over its full temperature range of

`Idaho Technologies The RapidCyclcr
`Technologies can amplify a 536 bp product in under 10
`minutes with a 30—cycle reaction. This speed is possible
`with a combination of photoheating, fan cooling, and thin~
`walled capillary reaction tubes. Thin—walled microcen»
`trifuge tubes may also be used. The reduced cycling times
`also result in reduced amounts of nonspecific products.
`Cycling times can be increased by programming in hold
`steps as desired. The unit carries a one~year parts and labor
`warranty. An optional buffer system allows finished sam-
`ples to be loaded directly onto a gel.
`
`Intelligent Bio-Instruments IBI offers the TC—1600
`high—thr0ughput cycler. It is capable of running 16 blocks

`accuracy. The lab need only btiy the single control unit and
`supplement it with the desired Alpha modules, such as
`dual, independently controlled blocks, microplate format,
`or a slide adapter for up to 16 slides. Thermal control is
`achieved by either block,
`in—sample probe, or calculated
`methods. Heating rates up to 3"C/second reduce reaction
`times while :0.4°C temperature homogeneity can be
`reached within 30 seconds at 90°C. The Hot Bonnet heat—
`ed lid controls pressure and height to accommodate vari—

`For high—throughput applications or busy labs,
`PTCv225 DNA Engine Tetrad may be the way to go. This
`allows up to eight separate blocks to run synchronous or
`independent reactions. A compact footprint is maintained
`by housing the power supply separately outside tie chassis,
`good for tight spaces or robotic applications. All ofthe fea-
`tures of the PTC—200 are present in this model as well.
`The PTC—10O cyclers incorporate a Peltier heating ele—

`after completion of the reaction. The compact, lightweight
`unit can be easily transported to different areas of the lab.
`Power failure protection resumes cycling after temporary
`power loss up to 12 hours.
`'lwo models are available for in rim work. The PTC-

`Tower block that can hold up to 16x2 slides. The PTC-
`l00—l6MS is a dedicated configuration of the PTC—l00
`series, able to hold up to 16 double— or sitigle—thickness
`slides. The sample block is made from oxygen—free copper
`milled from a single ingot and then plated in nickel for
`durability. Memory for 200 programs, power»failure pro-

`with a touchscreen display for easy interfacing. Multiple
`block options allow labs to run a variety of protocols.
`Personal memory cards let researchers store as many as
`l00 separate programs, and each program can be pass—
`word protected. Fast thermal cycling via Peltier heating
`allows a 30—cycle program to be completed in less than
`one hour to increase overall throughput. A stan

`The low—cost Progene cycler features a very small fei-
`print, high cooling and heating speed, wide thermal range,
`and a heated lid. Two intcrchaiigeable blocks are avail—
`able—one for 25 0.2—ml tubes and 5x5 inultiwell plates and
`the other for 20 0.5—ml tubes. A special feature is the abil~

`Whatman Biometra® \/Vliatman Biometra offers a wide
`range of cyclers to suit most needs. The T1 Therniocycl er
`is the flagship model, with -a number ofliigh-end features.
`Peltier heating brings fast ramping speeds of tip to 4°C/sec-
`ond heating and 3°C/second cooling. Remote control of up
`to eight units is possible with the available PC software. Lid
`pressure is variable with “smart—lid” technology. A large
`display facilitates programming with the intuitive interface.
`Microplates can be sealed during the first cycle with the
`specially developed microplates and heat—sealing film.

`and 96 models. Both feature a heated
`lid, NIST traceable accuracy (:0.5°C),
`fast heating and cooling rates (up to
`4°C/second), wide temperature range (0
`to 105C), parallel and serial ports, a
`two—ycar warranty, easy programming,
`and a small footprint. Model 64 holds 64

`microtiter plate or 96 0.Z—ml tubes.
`St. John Associates The BioOven III is
`one of the most unique cyclers in this
`review. Heating and cooling are done by circulating warm
`or cool air in a chamber that contains a rotating sample
`rack that can be easily switched to accommodate a variety
`of sample tubes, plates, and slides. Up to 236 0.S—inl tubes,
`10 microtiter plates, 30 slides, or hundreds of glass capil-
`lary tubes can be loaded in each run. The simplified menu-
`driven interface also includes a
`format
`feature, which
`optimizes conditions based on the type of sample bein
`run. A serial port allows users to program, format,
`access data from an external computer.
